Can paternity testing be performed without the mother participating?

Yes, Ningren paternity testing can be done without the mother participating.

Parentage analysis is basically a comparison of several genes between parents and a child. If we find the one of the child’s genes comes neither from its biological mother nor its alleged father (incompatible), and then the alleged father is excluded as the child’s biological father. If we find every tested gene of the child is compatible with the alleged father, we cannot rule out the alleged father is biological father. We, however, cannot be absolutely sure, because in this big world, it is possible that someone with no biological relationship with the child happens to have a set of genes compatible with the child. We need to further compare two probabilities: 1, the probability of the alleged father producing a child with the same genotype as the child being tested; 2, the probability of a random selected man producing a child with the same genotype as the child being tested. Based upon the two probabilities we can calculate the probability of alleged father being real father.

Simultaneously analyzing alleged father, child and biological mother can enhance our certainty about alleged father being real father (when not excluded). But due to the large number (15) of highly polymorphic genes chosen by Ningren for parentage analysis, even without biological mother’s sample, the calculated probability of alleged father being real one is usually quite large, larger than 99.99% (when not excluded) and exceed international requirement for parentage analysis. Therefore, under normal circumstance, Ningren does not need the mother’s DNA for paternity testing.
